Why a dedicated platform
General-purpose liquid handlers give you flexibility — and with it, every opportunity for protocols, reagents and operators to diverge. Evosep Lupo takes the opposite approach.
Methods are preset on the instrument, exactly as on Evosep LC systems, and the platform runs Evokits only. Consumables, protocol and instrument are co-designed and released together, so performance is characterised as a system rather than assembled from parts. The result is a workflow that behaves the same way on Monday and on Friday, in your lab and in a collaborator’s.

How it works
The starting point is a reduced and alkylated protein lysate — up to 1 µg in a maximum of 5 µl, with SDS at or below 0.5%. Everything downstream is handled by the system.

Performance
From application note AN-058A. K562 lysates processed with Evokit Digest, 1×96 (EV3802) on Evosep Lupo; Evotips analysed with 500 SPD and 200 SPD methods on Evosep Eno, with discovery analysis on an Orbitrap Astral and targeted analysis on a Waters Xevo TQ-Absolute XR.

Challenged with real-world contaminants. A complex bone tissue matrix (1 µg starting material, 30% of digest loaded on Evotip, n = 8 per matrix) spiked with 4 M GdnHCl, 20% glycerol, 0.2 M glycine, 0.5 M NaCl, 2% NP40, 0.5% SDS, 0.5 M Tris, 1% Triton X-100, 2% Tween 20 and 4 M urea. Quality control and traceability
Two complementary controls ship inside the kit and execute as part of the routine workflow, so quality assessment adds no operational complexity.
Process Control Digest — 1 µg K562 in 0.5% SDS, processed twice per plate in positions A12 and B12, monitoring the full sample preparation workflow. The proposed LC-MS run order places them as injections 3 and 4.
System Suitability Control — reporter peptides that verify LC-MS performance over time by tracking retention time, peak shape, peak width and signal intensity. Chromatographic measures determine QC quality and can be assessed in real time.
Default run order of QC samples. Process control samples monitor the full sample preparation workflow; system suitability controls verify LC-MS performance over time.
The output view lists every experiment with kit, loading percentage and sample count, including QC samples — with report, copy and export actions. Sequence lists export as a tailored .csv for Thermo, Bruker, Waters, SCIEX and Agilent software.

Evokit Digest is optimised to work in the presence of common lysis buffer components at these concentrations. SDS is removed during the workflow and is not loaded onto the Evotip.

Evokits combine reagents, labware, Evotips and process controls with a predefined protocol, packaged for direct use in Evosep Lupo. Data matrix codes on all components carry protocol identity and execution detail, and each kit ships with a certificate of analysis.

Frequently asked
The complete Evokit is processed at once — once reagents are pierced and dissolved, stability cannot be controlled over longer periods. Fewer than 96 samples can be processed, but the instrument will still handle the full plate and consume reagents accordingly. We recommend pooling to prepare as many samples as possible per run.
No. Methods are preset on the instrument, as with Evosep LC systems. The platform is standardized and works with Evokits only — this is what allows the performance of the complete workflow to be guaranteed.
No. Evokits are not supported on other liquid handlers, and Evosep Lupo can only run Evokits manufactured by Evosep. Kits can be purchased without the instrument, but they are designed for automation and can only be processed on Evosep Lupo.
No. The endpoint of every Evokit is a loaded, ready-to-analyse Evotip. The system cannot load samples prepared manually or on a different liquid handler.
To reduce hands-on time and increase consistency for every sample. Automation delivers the same result each time and, with standardized Evokits, requires no optimisation — alongside built-in traceability, minimal required experience and seamless integration with Evosep LC.
Digestion runs at ambient temperature for 4 hours. Digestion efficiency is stable across temperatures and has been tested down to 20 °C.
Samples should be aliquoted into 4 × 96-well plates prior to preparation. The instrument processes up to four plates in parallel — 96, 192 or 384 samples per run.
No. One loading amount is used across a plate.
No, peptide quantification is not part of the workflow. We recommend a BCA assay to normalise protein input before the run.
Chromatographic measures are used to determine the quality of the QC samples, and this assessment can be made in real time.
The initial Evokit portfolio covers digestion and quality control, with a clear path toward application-specific kits. The ambition is to enable enrichment workflows as well.
Evosep will continue to service the instrument and Evokits. Performance Service Contracts are available to protect uptime and analysis productivity.
